[Medoc-CPM]Tapentadol potentiates descending pain inhibition in chronic pain patients with diabetic

This study investigates Tapentadol’s analgesic effect on diabetic painful neuropathy (DPN) patients and demonstrates that the pain relief is coupled to reactivation of descending inhibitory pain pathways, assessed by CPM test

-Tapentadol - m-opioid receptor agonist, inhibits neuronal reuptake of norepinephrine
-Performed on 24 DPN patients ? 12 with placebo treatment and 12 with tapentadol treatment.
-Before treatment ? none of the patients demonstrated significant CPM responses
-After 4 weeks of treatment, CPM was significantly activated by tapentadol
-CPM activation was correlated with significant analgesic responses

“Our results show that patients with DPN that

display absent CPM responses benefit from tapentadol causing

pain relief coupled to (re)activation of descending inhibitory

pain pathways”
